Spurs’ 61,000 capacity arena is yet to be completed, but will be hosting the first of the games.
In Week 6 of the season, Seattle Seahawks will take on Oakland Raiders (Sunday October 14).
Philadelphia Eagles play Jacksonville Jaguars, while Tennessee Titans take on Los Angeles Chargers in the other two matches at Wembley, to be played on October 21 and 28.
Last year saw four matches take place in London as the NFL celebrated the tenth anniversary of the first games in the UK.
The NFL and Spurs announced in 2015 that games would be held annually at the football club’s new stadium, with rumours also of a London based franchise.
